Independent regulation of the nuclear RNA polymerases I and II during the yeast cell cycle.

چکیده
The activities of the nuclear RNA polymerases I and II have been investigated in cells of Sacharomyces cerevisiae at G(1), S, early G(2), and late G(2) stages of the cell cycle. The results show that the ratio of the activities of the two enzymes is different at these four stages, indicating that the two RNA polymerases are regulated independently during the cell cycle. The specific activity of the RNA polymerase I remains constant but that of RNA polymerase II increases sharply in cells at the beginning of the G(2) phase. These results are compatible with a pattern of continuous synthesis of the RNA polymerase I and step-wise synthesis of the RNA polymerase II during the yeast cell cycle.

Subunits shared by eukaryotic nuclear RNA polymerases.
RNA polymerases I, II, and III share three subunits that are immunologically and biochemically indistinguishable. The Saccharomyces cerevisiae genes that encode these subunits (RPB5, RPB6, and RPB8) were isolated and sequenced, and their transcriptional start sites were deduced. RPB5 encodes a 25-kD protein, RPB6, an 18-kD protein, and RPB8, a 16-kD protein. These genes are single copy, reside ...

Background: Progesterone is indispensable for differentiation of human endometrial stromal cells (HESCs) into decidual cells, a process that critically controls embryo implantation. However, HESCs also abundantly express androgen receptors (AR), yet the role of this member of the superfamily of ligand-dependent transcription factors in the decidual process remains poorly elucidated. Materials a...
متن کاملQuantitative conservation of chromatin-bound RNA polymerases I and II in mitosis. Implications for chromosome structure
RNA synthesis almost ceases in mitosis. It is ambiguous whether this temporal, negative control of RNA synthesis is solely because of the nature of chromosomes per se, (i.e., their condensed state), or to a physical loss of RNA polymerases along with other nuclear proteins which have been shown to pass into the cytoplasm in mitosis, or to their combined feature. Aside from such regulatory consi...
متن کاملThe TATA-binding protein is required for transcription by all three nuclear RNA polymerases in yeast cells.
Using temperature- and proteolytically sensitive derivatives to inactivate the function of the yeast TATA-binding protein (TBP) in vivo, we investigated the requirement of TBP for transcription by the three nuclear RNA polymerases in yeast cells. TBP is required for RNA polymerase II (pol II) transcription from promoters containing conventional TATA elements as well as functionally distinct pro...
متن کاملTranscription in yeast: alpha-amanitin sensitivity and other properties which distinguish between RNA polymerases I and III.
Three peaks of DNA-dependent RNA polymerase (RNA nucleotidyltransferase) activity are resolved by chromatography of a sonicated yeast cell extract on DEAE-Sephadex. The enzymes, which are named RNA polymerases I, II, and III in order of elution, show similar catalytic properties to the vertebrate class I, class II, and class III RNA polymerases, respectively. Yeast RNA polymerase III is readily...
